Overseer was a title used by the Androssi. An Overseer often commanded a salvage team, and oversaw all actions carried out. Androssi that had carried out a brave or profitable action in the team were often promoted to Overseer.
An Overseer was the leader of a Vault-Tec Vault and according to Fallout 3 Survival Guide a Vault-Tec official. The Overseers were often aware of the Vaults being an experiment and helped coordinate it. The Overseer served many purposes from Mayor to (in some Vaults) a sort of god.
According to the letter from Dr. Braun, in the Vault 101 Overseer terminal, if the overseer disclosed the contents of the sealed orders or deviated from the experiment, it can be grounds for termination of employment from Vault-Tec. The Overseers receive their sealed orders from Dr. Braun, the terminal from Vault 101 contains an article about the GECK, which is described as the most advanced device known to science. Do to "cost concerns" not all Vaults will be equipped with a GECK.

The Overseer (Also known by her real name, Jill) is an NPC who runs the Sawmill Jobs minigame. Players need to talk to her after completing the job they were given to receive the extra exp. When you talk to her she says "Yes? What do you want? I'm very busy!" Which is a bit of a strange comment because she doesn't really seem to be doing much!
She stops players trying to leave with planks or logs in their inventory, confiscating them.
